I'm doing it for a friend. What's the best way to not lose any files ie pics/music etc?

Do I 'backup' on iTunes and THEN update iTunes and update the iOS? How to I check to make sure all music etc is backed up?

just back it up in iTunes before updating to 6. It's easy, just right click on the iPhone under devices in the left bar and click backup. hope it helps Everything on the iPhone now will be on the iPhone once the upgrade is complete And you restored it to the iTunes backup

Do I need to sync its photos and music and videos? Or will it automatically save them on the backup? Thanks

Also- is it better to update the iTunes software on PC before or after I backup
 
Before it updates, it auto back up everything, provided that you tell itunes to backup photos, and anything that there. Like upgrading a Mac operating system, files aren't touch when the OS is upgraded.
 
Be careful. iTunes won't backup any music, photos or videos. All you can sync to your computer is the content of the camera roll (it contains the photos you took yourself, pictures you saved from other applications, the Internet etc). But: music, movies and photo libraries (from iPhoto etc) your friend copied to the phone at some point will not be synced to your computer. It works sort of one way only...the best would be that your friend syncs the iPhone to the computer they used to get all the music and stuff onto the phone in the first place.
