I have a 17" 2010 MacBook Pro. I have a card reader expansion card in the ExpressCard/34 slot that I never use, and I never use my optical drive.

Any suggestions for what I could utilise them for? eSATA? USB3?

I did toy with the idea of putting an SSD in place of the optical drive, but I already have a 750GB SeaGate hybrid drive as my HD.

SSD in main bay and your 750GB HDD in optical for extra storage.

I use my 34 slot for 2 USB3.0 ports.
Similar here -- SSD in main bay and 500GB HDD in optibay in a late '08 15" MBP.

I have an SD card reader in the 34 slot -- very handy for transfer of photos from camera SD card.

Also, you can put your Superdrive into an external enclosure. I went with MCE electronic's $30 optibay HDD adapter and $20 Superdrive external enclosure package - $56.xx with shipping.
