What version of iTunes do you have installed? Does your iPod show up in iTunes when it's connected via the USB cable?

Are Applications showing in the left sidebar in iTunes?

If not, open iTunes Preferences > General and click the checkbox next to Applications and see if it shows up. This may then force the applications tab to show when the touch is connected.
