I'm planning on the next refreshed 15" Macbook Pro now. I plan on putting it on my night stand, playing music through external speakers, keeping it plugged in most of the time. However there will be times when I do take it off the charge to take around. Will this do harm to the batteries life?

As long as you run it on battery every few days, you'll be fine.

This should answer most, if not all, of your battery questions: Apple Notebook Battery FAQ

Exactly. Make sure you keep a good mix of battery usage and it shouldn't be a problem.
