In the past, I have purchased MacOS and with 1 copy I have installed it on both my MBP and my wife's MB or will it require separate purchases?

Also she is using Leopard. My understanding is that I need to upgrade her computer to Snow Leopard before I can install Lion?

You can install Lion on any machine that you authorize with your Apple ID, so you only need the one copy.

I'm not sure about how to install over Leopard. You might try some tricks people have talked about that involve right-clicking the installer app, selecting "Show Package Contents", and then find the installer bits inside there to copy over to Leopard. See if that works. Or someone else can supply a better answer.
