Is there a way to lock many files at once?
I'm aware of that doing a "Get info" will allow me to change the lock status, but with hundreds of files in a folder this isn't very practical.
 
Open the folder - highlight whichever files you want to lock or press Command A to highlight all. Hold down Command & Option keys and press 'i' key. This will open the multiple Get info window. Check the Lock box. Now - whichever files have been checked will be locked.

Wow, I didn't know about CMD-OPT which results in "Show inspector" for multiple items. Very useful. Thanks!
 
……..And, just in case you want to do it in Terminal.

1. Open a Terminal window

2. Type in: sudo chflags uchg (with a space at the end)

3. Drag the files you want to lock into the Terminal window

To do the same in reverse use: sudo chflags nouchg
