So i have 4GB of ram at the moment in my MacBook Pro Mid 2012 .It has 2 memory(ram) slots If i buy this ram (8GB) will i be able to take out one 2GB module and insert a 8GB to replace the 2GB making The total ram 10GB? Yes and yes. As an alternative, you could get something like this 8GB kit (4x2) and replace both modules, giving you a matched pair.

Will two different ram manufacturers work?
The brand makes no difference, as long as the specs are correct. There is a slight performance benefit in using matched pairs of RAM. If you don't need more than 8GB, you would get slightly better performance with the matched pair than with the 2GB and 8GB modules.

im not from USA or Uk im from South Africa so the shop where im getting it from is cheaper for one 8GB module and i can always get another 8gb to match if i want 16GB ... But the question now is will the ram that came with my mac (4GB)-2 Of 2Gb modules .. If i take out one 2gb module and put a 8gb in will wit work ? since the rams are both different makes - manufacturers

As I already stated, yes, it will work and it doesn't matter if the brands are different, as long as the specs match.
