When I customise the toolbar of Mac Mail, adding the facilities I want (such as 'Move' and 'Go On/Offline' etc.) everything is ok until I close the app. When I open it again, all customisation has disappeared.

Any ideas, anybody?

Quit Mail
Rename (or delete) the user preference file for Mail.app, located in ~/Library/Preferences/com.apple.mail.plist
Restart Mail.

Note this will effectively reset *all* your user preferences

Thank you - but it's still the same... doesn't even retain the 'text & icons' setting :(

Are you on Mavericks? Mavericks caches plist files and plist deletes won't stick unless you restart.

So quit Mail and delete the plist like RemarkabLee said. Then restart to flush the plist cache and now start Mail to go to a default plist.
