I didn't know the 2010 21.5" iMac had access doors? I seem to remember that a screen had to be removed below the display before the memory modules were visible. Anyway, go to www.ifixit.com and find out for yourself.
 
I am pretty sure the 2010 27-inch has a longer access door, compared to the 21.5-inch.
Those both have access doors on the bottom edge.
When Apple went to the thin cases in 2012, then you got the hatch on the back of the 27-inch, and the 21.5-inch lost easy RAM access.

21.5” 2011s & earlier had RAM accessible on the bottom of the screen with a screwdriver. In the 2012s they removed the modular RAM entirely for the 21.5”.

Nope... on the bottom there is a plate you unscrew to access memory... just upgraded from 4 to 12 gb and replaced the old seagate 7200 rpm drive with a SSD... :).

Yeah, I mislabeled it as a screen but it is in fact a plate with holes and screwed to the bottom of the chassis below the display. The plate on the 21.5" model will not fit the 27" model.
