I would like to sell my curent computer before buying the new one. Is it possible to use Migration Assistant with an external hard drive as a go-between, instead of going from computer-to-computer?

yes, either clone your Mac to an external hard drive or use a Time Machine backup and select either from Migration Assistant.

Thanks!

OK, so if I use Time Machine, can I do it with an eSata drive as the middleman? Or does it have to be a Firewire drive?

Time Machine can use any hard drive that appears as a volume and is HFS+ formatted.

Right, but what I meant was if I use Time Machine, can I then use Migration Assistant, targeting that disk that was backed up with Time Machine. Or does it have to be a FireWire drive? (I read something about FireWire exclusively being able to work in "Target" mode or something like that.)

no it doesnt have to be Firewire. it can be any connection USB, eSATA, Firewire. Target Disk Mode can only use Firewire.
