Yes you will, I'm guessing this is just useful if you're from out of the country and don't have a shipping address, or you live near the border of a state with lower sales tax.
It's treated like an instore purchase so if you live in DC and order online for pickup in Delaware then you don't pay any sales tax. Obviously you pay DC use tax.
Yes you can order in store. I had cash and didn't want to use a credit card. I went to the store and they put the cash on an Apple Store Card (not iTunes Card) and I was able to do everything at the store on their computer. I set it up to be delivered to the store for pickup. Didn't have any problems.
