Yes you can do that as long as you have the cellular version of the watch. I think a couple months ago Spotify updated their app to stream on the watch

The Apple watch can stream Apple Music over cellular or WiFi. Spotify appears to be a different matter and currently requires a connection to the iPhone. Check out this search: Can Apple watch stream Spotify over wifi?
