I have 1 airport extreme at the moment. It is connected to the modem in my kitchen now my home office is roughly 20ft away would adding a second aebs to my network and connecting my computer to it through Ethernet rather than wifi make the connection faster on my computer?
 
I'm guessing that your computer has Wi-Fi built in. If so, it would be the same speed connecting it through the AirPort Express as it would be connecting to the Extreme network directly. The Express would only help if there was a greater distance.

No, it would create much more latency issues. Just run a CAT5 or CAT6 cable to your office. That or get an Airport Express station to extend your wireless network. Both will still be the same speeds (wireless vs wired). However, a much more responsive connection is achieved wired.
