I have a 6th Gen APE which is our “main” router on the 2nd floor and a 5th Gen APE on the 1st floor we are using as to access point for our A/V equipment and to extend the wifi network. Is there a way to find out the connection speed between the routers using the AirPort Utility?

Yes. Select your base station and hover over the connection quality.
