iPod touch Syncing iPod after reinstalling iTunes

Had to install a new HD on my PC. I then installed iTunes 10 on the PC, set up my account again and purchased one song.

I then connected my iPod Touch to iTunes. When I attempt to sync, iTunes wants to replace everything on my iPod with the song I just downloaded!! How do I backup and save the contents of my iPod?

This is my grandson's iPod so any help would be greatly appreciated.
