Tagging files for improved search results?

Is there a way to tag files to improve search accuracy? What does Spotlight use when it searches files on a Mac? Just the title? Anything else?

When you do Get Info on a file, there will be a box near the top called Spotlight Comments. Presumably Spotlight uses this (it should do with a name like that!) although I've never tried it myself.
