This biggest question mark about the new Watches is whether you will be able to order online with in store pickup on launch day tomorrow Fri. Sep. 18. Right now that option is grayed out on website / apple store app but no word if inventory will be loaded and this feature available for tomorrow. Any one with any info on this?

I believe the option will be available sometime early tomorrow morning, once stores get their stock and accurate stock info goes live.
Some Best Buy’s will have them in stock tomorrow, I’ve seen a few Apple watches available for in store pick up tomorrow morning on their website.
 
I did see that Best Buy has some for pickup tomorrow but they only stock specific case/band combinations. For example if you want blue watch they only offer with blue sport loop (not even the blue solo band). Given the numerous sizes for solo band I doubt you'll see those bundled at Best Buy.
 
Stores in my part of the world will have stock for walk in but not reserve and pick up.
 
I spoke to an Apple customer service agent for Century City store in LA. She said that the stores will get the watches on Monday. You can preorder to pick up on Sunday. Interestingly, you cannot make an appt to shop with a specialist in LA this weekend, but they just released appointments for Monday.
