I have an iMac and a MacBook Pro. If I buy a large usb hard drive and connect it to the iMac can I back the MBP up to it over the wireless network?

yes, you can back up your Macbook Pro to the iMac's connected drive.
The second poster is completely wrong about the Airport Extreme's backup capabilities, too. I'm backing up to my Airport network drive as I type this.
