I have traffic conditions and frequents locations set to on. I got the 6S on 10/5.

If you are referring to how long it will take before the iPhone will start showing you this information, I found it to be roughly 2 weeks. The iPhone needs to learn your patterns, i.e.. what time you leave for work each day, when your appointments are in your calendar, etc. I placed my work schedule as a recurring event into my calendar to try to help it understand my patterns.

Does adding it to the calendar speed up the process?
 
Mine isn't showing yet either. It's been about 2 weeks so hopefully it'll start soon. Although I don't rely on it much since my commute is all surface street with damn traffic lights :)

Mine took a couple weeks to learn too. Just started getting traffic info as a notification within the last week or so.

I got my iPhone on launch day.

It seems like my iPhone 6 used to tell me about traffic updates on my commute more often but I rarely get updates any more. Did they enhance this feature with iOS 9, or am I confusing it with something else?

I agree with this.
 
Had my 6S Plus since launch, and just started getting the traffic predictions again yesterday (again, because I had them on my 6 Plus).
 
Yes I get traffic predictions, and I if I watch my iPhone siri tells me the drive time before I leave for class.
