I had my phone on 4.0.1, then I updated to 4.1, then I went back to 4.0.1 can my phone still be unlocked or no?

NO. Updating firmware also updates baseband, but downgrading firmware won't downgrade the baseband. You will have to wait for 5.12.04 baseband unlock. That's why it's even more important not to update if you need to unlock since there is no way to go back.

How did you update to iOS 4.1? Did you use Tiny Umbrella and iTunes or just iTunes? What kind of iPhone do you have?

Most likely no.
Unless you're talking about an iPhone 4 and you had your hosts file edited or TU running in the back while you updated thru iTunes.
