Every time you update firmwares, you will need to re-jailbreak and unlock. Backup your iPhone, and run the appropriate version of redsnow.

You can upgrade to 3.0.1 in iTunes if you REALLY want to, and then run redsn0w 0.8 and point it to the 3.0 IPSW and you will have a freshly jailbroken 3.0.1 phone.
