I got a few USB hubs and I was wondering how much 1 USB port 'can take' ?

Or do you just have to divide the USB transfer speed by the number of devices using the port at that time?

And if so, will devices plugged in (but not being used at the time) lower the USB transfer speed then as well?

Max USB capacity is 127 devices on a single bus. Powered/bus powered, device draw, device type, cable quality & length, all these will affect usability and available throughput.

I'm running as many as ten or so from the MacBook when I'm in "desktop" mode. On the road, generally no more than two. I've never gotten to the point where devices have issues, and certainly not measurable (much less noticeable) throughput hits.

This includes mice, trackballs, keyboards, cameras, scanners, portable HDD, etc. I have a 7-port stackable Belkin powered hub I use with great success.
