Hi,

My iPhone 4's + and - are very inconsistent. The + button is very tough and actually difficult to depress, and the - button is very clickly and easy to depress. Would this be considered manufacture tolerance? The + button is too tough for my liking and some could even say it almost feels like it's a little bit more depressed into the phone than the - button. What do you think?

OCD. LOL.
If its not broken, dont fix it. Think of it this way. Its better that the volume button be more "solid" and "hard to depress" than have volume buttons that are loose and rattle-ly like my phone buttons.

From the sound of it, the OP's Volume Minus button is somewhat loose and rattling.
