You need Show Lep or Lion.

also check your settings in both itunes and on the phone. only other requirement is that it needs to be on the same wifi network, running the latest itunes 5.0 & Lion/Snow Lep.

I think you just need iTunes 10.5 and have your Mac/iPhone/iPad/iPod touch on the same WiFi network. It says you need to have it plugged in but I've been wirelessly syncing my iPhone without it plugged in

Only works with OSX 10.6 and 10.7 - all current Windows versions work though.

Another thread just revealed that it requires Snow Leopard or Lion. It won't function under Leopard, which is what I am running.

A little off topic but so save starting a new thread:

It seems you don't have to be plugged into the mains in order to sync via wifi like everywhere/one says, I just sync'd my iPhone 4 over wifi without being plugged into a charger :)

I read a blog somewhere that automatic syncing starts when you plug to charge your iphone, but you can start manual syncing at any time.
