When I'm backing up using Time Machine and I knew it will take quite awhile (est 3 to 4 hrs as it has been a long time since I previously back up), and my normal setting for computer sleep is 15 mins, will it stop backing up in 15 mins? Or I have to adjust the sleep longer?

I too would like to know the answer to this and also to know what other actions can take place during sleep mode.
 
Depends...on your OS and hardware. Older Macs may sleep, but with Power Nap on recent OSes.....TM will still work in the background (along with lots of other stuff). Details here.

For older machines, you could schedule the machine to wake from sleep, or restart. It may take an hour or so for TM to kick off. You could use Automator to schedule TM to run immediately.

Or, you could simply set the Mac so that only the display sleeps. Leave the OS and HD running, problem solved.
