I have a Linksys E2500 router as the only wireless access point in my home. I'd like to make a separate network using an AirPort Express by connecting the Express to the Linksys via an ethernet cable. Will this work?

Yes that will work.
(my opinion) it will work best if you make the A/Expr a different network name, diff. channel and IP subnet.

Yes. It will work fine. Remember to use identical settings if you want clients to roam between APs.
