It makes zero sense to only offer the upgrade program for one day. The whole purpose of this program is to lock people into iPhones, making it easier and more affordable (for those who have trouble purchasing in full up front) to get an iPhone every year, which just makes people more invested and enmeshed in Apple's ecosystem. Why on earth would they only offer it for one day?

All right, I just got some info which answered my question as well as those in this thread prior to mine:

-You can purchase an iPhone at any time from an Apple Store using the iPhone Upgrade Program. You don't need an appointment. It is basically another payment option.
-This includes in-stock iPhones on Sept. 25.
-The reservations are currently closed. However, starting Sept. 26 at 8 a.m., they will reopen online and you can reserve a phone for that day. It will be updated each day at 8 a.m.
-So, as already said here, the iPhone Upgrade Program will be offered beyond launch day.

Hope that helps everyone out!
