Do you mean the web version of Plex will work with the built in PIP mode in macOS Sierra? You need adBlock extension in order to do this? Could someone please explain more on how to get this working? I am using u-block and I don't know what this means 'app.plex.tv##button.video-overlay-btn'

am I suppose to type this somewhere?

FYI, you can use PiP with any HTML5 video, make sure you have developer tools turned on in safari settings. Then go to the develop menu and click disable styles. This will remove any overlays and customizations a website adds to the video. Scroll down until you find the video and right click - enter Pip, then go back to develop and re enable styles.

A little work around until more sites add built in functionality to their ****** UIs

I haven't tried that method, but on Netflix, I was able to use PIP by disabling JavaScript so that you can right-click, and then once in PIP mode, you can re-enable JavaScript
