Signature in Mail

I wonder whether one can make the signature optional -- to select if she/he wants it or not on per-message basis?

If you are speaking of the stock Mail app, yes. If you are speaking of Outlook or some other mail app, most that I have seen make the signature optional.

Other than deleting the signature in an outgoing iOS Mail message I'm not sure how BasicGreatGuy is accomplishing this.

Pretty sure you can't.

If you don't mind a one-line signature...something like:

John Doe • Doe Enterprises • (800) 555-1212​

you could use a "Text Replacement" shortcut.
