My wife just bought 3 Apple watches for friends of ours directly over the counter at an Apple store in Chicago, so I'd say yes to your question.
All shops might not have all models, but most will have a decent selection to choose from by now.

I think even the Space Black 42mm with Space Black Link Bracelet is now available in most Apple retail stores, but I would look online at the store availability listing on the day your brother is planning to go to the store just to be sure......
still no apple watches at best buy in wichita ks but I noticed a store in Tulsa and OKC now have them. Of course those stores have apple stores so there would be stock there most likely. I did notice that best buy already had an open box apple watch and it was priced $19 cheaper than the "new" ones.
Saw the Watch for the first time in person yesterday. First impression- cool watch (as in, it looks like a nice piece of jewelry/ time piece). Second impression- it is much smaller than any images can convey. I was really surprised when I started looking for the larger version, when in fact I was already looking at it! I was also surprised to see about 6 edition models on display (under glass of course). Both my sister and I agreed immediately though that the stainless steel model would be the only one we'd be interested in buying. The sport just looked mediocre for some reason. Will wait for the second generation and more independence from iPhone before buying though.
He should be able to get one without having to reservation. Apple got rid of the reservation system for the apple watches as they are readily available at the apple store. However, he can still visit apple.com and he can purchase it and have the option to pick up in store.
Bought a 38mm stainless steel brown leather modern buckle model over the weekend for someone. My local Apple Store had them in stock, along with classic buckles and Milanese loops.
