Umm...no you can't, not so that the reply address defaults to whatever you want *and* displays as such on all mail clients. And I wish people would stop spreading this misinformation.
Yes, you can forward any address (directly or through gmail) to your me.com address. Yes, you can change the smtp server on your iphone to get it to seem as if you're replying from a different address.
HOWEVER, when said mail is received on certain mail clients/services (e.g. Hotmail), the recipient will see something to the effect of
"From: yourcustomemail@customemail.com" brought to you courtesy of Gmail." (or something like that - it's been a while since I checked it out myself)
On most mail clients/services it looks fine - like it comes directly from your custom email address. But some - enough in my book - display the ungainly additional information, information which looks unprofessional. And since looking professional is one of the key reasons to having a custom domain email, it pretty much defeats the purpose.
