Jacob and Esau are twin brothers in the Bible. Esau was the eldest and Jacob popped out holding Esau's ankle. You can read more about their story here, but as of right now, I don't see any connection with Lost. Maybe that'll change, but I doubt it. I don't think MiB has ever been named.
I don't know... Maybe not the biblical version (still some, see below), but if you scroll down to the Book of Jubilees, you can definitely see some parallels.

This is pretty clearly attached to Lost, if you replace the father with their 'Mother'.
Book of Jubilees said:However, after the death of Isaac (again, replace with mother), the sons of Esau convince their father to lead them, and hired mercenaries (Ben Linus), against Jacob in order to kill Jacob and seize his wealth (this wealth would be the ability to leave the island and have the 'source').
Again, pretty clearly attached to Lost, and if you say that Ben was a hired mercenary who did MiB's bidding for him.
Book of Jubilees said:The sons of Jacob (Jack, and others it appears) then defeat the rest of the attackers despite overwhelming odds. Some of the sons of Esau are spared, but they are sworn to serve and pay fealty to Jacob.
Could this be the ending? If you say that Jack is the figurative 'son' of Jacob, then this works! It also makes sense that some of Esau's figurative sons, such as Sawyer, are redeemed and help Jack in the end protect the island.
This could also show why they were reluctant to show what MiB's name was! They must have given him a name, because he would know that it was odd that Jacob had a name, and he didn't! So, it's pretty clear they deliberately hid it! If Lost ends this way, I deserve immediate bragging rights!

P.S. The Biblical version (21:7) also says how Jacob used deception and trickery to get his father's birthright (this could be seen as protection of the source by his mother).
